Constitutively Expressed QseC
Figure 1: Sequence overview of BBa K4010011.
The two-component Quorum Sensing E. coli regulator B and C system (QseBC) is responsible for inducing flagella expression and therefore bacteria motility (Clark et al., 2006). QseC is a histidine sensor kinase that acts as a membrane-bound adrenergic receptor, responding to both microbiota-derived autoinducer-3 and the host hormonal stress signals epinephrine and norepinephrine (Clark et al., 2006; Rooks et al., 2017; Sperandio et al., 2002). Upon activation, QseC autophosphorylates and transfers a phosphate to QseB, which is the response regulator in this signaling cascade (Clark et al., 2006; Rooks et al., 2017). Phosphorylated QseB directly binds to both the high and low affinity sites of the flhDC promoter region, leading to transcription of downstream flagellum regulator genes (Clark & Sperandio, 2005). Since both sufficient quantity and phosphorylation of QseB are required for full activation of flagella regulon, the bacteria ensures the presence of survivable conditions with ample resources in host before flhDC transcription (Hughes et al., 2009).
